ABSTRACT
SRI NURUL ‘AINI. J2B003098. CLONING GENE xyn ENCODING XYLANASE ENZYMES FROM Geobacillus stearothermophillus IN TO Escherichia coli DH5α. GENETIC LABORATORY OF DIPONEGORO UNIVERCITY & CBRG (Carbohydrat Bioengineering Research Group) RESEARCH CENTER FOR BIOTECHNOLOGY - INDONESIAN INSTITUTE OF SCIENCE. TO BE LEADED BY HERMIN PANCASAKTI KUSUMANINGRUM REJEKI SITI FERNIAH AND BUDI SAKSONO.
Xylan has an important role in hydrolysis xylose from xylan. Xylanase have important applications in rare sugar, pulp and paper industry. Tey can also be used to increase the digestibility of animal feed stock and in the baking and brewing industries. A discovery has been done to get gene xyn encoding xylanase enzyme from Geobacillus stearothermophilus. Amplification of gene xyn has been done by PCR RTG (Ready To Go) using primer xynF and xynR that designed to get a 1239 base pairs gene as described in NCBI GeneBank. This fragment has been succesfully cloned into a plasmid vector pGEMT-easy giving a pGEMTxyn wich is transformed into Escherichia coli DH5α. Sequence analysis of recombinant plasmid pGEMTxyn showed that the gene was exactly xyn from G. stearothermophillus. Gene xyn was then subcloned into expression plasmid pET-21b(+) and introduced in E. coli BL21 for protein expression study.
